چکیده
This pioneering work reports successful androgenic plant development via embryogenesis from microspore calluses in anther cultures and estimation of bioactive metabolites vitro regenerants parent (control) Cambod tea, Camellia assamica ssp. lasiocalyx (Planch MS) cultivar TV19. Anthers bearing microspores at early-to-late uni-nucleate stage were selected to initiate androgenesis. A pre-treatment 5 °C for days the dark was most effective profusely growing white callusing within 10 weeks culture on MS medium (6% sucrose) supplemented with high cytokinin/auxin ratio maintained by 6-benzylaminopurine (BAP) 2,4-dichlorophenxoyacetic acid. Nodular structures callus surface differentiated into embryos. Further developement embryos occurred but, ten times reduced concentration growth regulators additives. Germination complete plantlets achieved when major salts half (½ augmented BAP, GA3 IBA along glutamine serine. Cytological examination root-tip cells revealed that regenerated haploids (2n = x 15), which further confirmed through flow cytometry. The hot-water extracts haploid calluses, field-grown donor utilized quantification (+)-catechin, (−)-epicatechin, (−)-epigallocatechin gallate, caffeine theophylline. Our findings metabolite profile is comparable mother plant, thereby presenting them as potential source genome duplication genetically stable homozygous pure breeding lines. first report out-breeding tree, tea. It’s a significant achievement towards generating lines, impossible using conventional methods. Haploids showed consistent production.

Somatic Embryogenesis and Plant Regeneration from Embryonic Axes and Cotyledons Explants of Tea (Camellia sinenesis L.)
In the present study, 2, 4-dichlorophenoxyacetic acid (2, 4-D) was assessed individually for its effectiveness to induce somatic embryogenesis in tea (Camellia sinenesis L.). Embryonic axes and cotyledons explants were dissected from the seeds. Explants were cultured on Murashige and Skoog (MS) medium containing 0, 1 and 5 mM 2, 4-D alone for embryonic axes and 0, 1 and 5 mM 2, 4-D along with 0...
متن کاملComparative Transcriptome Analysis of Chinary, Assamica and Cambod tea (Camellia sinensis) Types during Development and Seasonal Variation using RNA-seq Technology
Tea quality and yield is influenced by various factors including developmental tissue, seasonal variation and cultivar type. Here, the molecular basis of these factors was investigated in three tea cultivars namely, Him Sphurti (H), TV23 (T), and UPASI-9 (U) using RNA-seq. Seasonal variation in these cultivars was studied during active (A), mid-dormant (MD), dormant (D) and mid-active (MA) stag...

PURPOSE This study aimed to evaluate the preventive effects of Camellia sinensis var. assamica (CSVA) on diabetic nephropathy in in vitro and in vivo models. MATERIALS AND METHODS MDCK cells were incubated with 1 mM of oxalate with or without different concentrations of CSVA, then MTT and malondialdehyde (MDA) assays were performed to investigate the preventive effects of CSVA on oxalate-indu...

متن کاملGenetic Variation of Flavonols Quercetin, Myricetin, and Kaempferol in the Sri Lankan Tea (Camellia sinensis L.) and Their Health-Promoting Aspects
Flavonol glycosides in tea leaves have been quantified as aglycones, quercetin, myricetin, and kaempferol. Occurrence of the said compounds was reported in fruits and vegetable for a long time in association with the antioxidant potential. However, data on flavonols in tea were scanty and, hence, this study aims to envisage the flavonol content in a representative pool of accessions present in ...
